Spread Plate Technique The spread plate method is a technique to plate a liquid sample containing bacteria so that the bacteria are easy to count and isolate Procedure 1. Make a dilution series from a sample. 2. Pipette out 0.1 ml from the appropriate desired dilution series onto the center of the surface of an agar plate. 3. Dip the L-shaped glass spreader into alcohol. 4. Flame the glass spreader (hockey stick) over a Bunsen burner. 5. Spread the sample evenly over the surface of agar using the sterile glass spreader, carefully rotating the Petri dish underneath at the same time. 6. Incubate the plate at 37°C for 24 hours. Streak Plate Method Streaking is rapid and ideally a simple process of isolation dilution.The technique is done by diluting a comparatively large concentration of bacteria to a smaller concentration. Procedure • A small amount of sample is placed on the side of the agar plate (either with a swab, or as a drop from an inoculating loop • A sterile loop is then used to spread the bacteria out in one direction from the initial site of inoculation. • This is done by moving the loop from side to side, passing through the initial site. • The loop is then sterilized (by flaming) again and the first streaks are then spread out themselves. Serial dilution, Serial dilution as the name suggests, is a series of sequential dilutions that are performed to convert a dense solution into a more usable concentration. Procedure 1. The sample/culture is taken in a test tube and six test tubes, each with 9 ml of sterile diluents, which can either be distilled water or 0.9% saline, are taken. 2. A sterile pipette is taken. 3. 1 ml of properly mixed sample/culture is drawn into the pipette. 4. The sample is then added to the first tube to make the total volume of 10 ml. This provides an initial dilution of 10-1. 5. The dilution is thoroughly mixed by emptying and filling the pipette several times. 6. The pipette tip is discarded, and a new pipette tip is attached to the pipette. 7. Now, 1 ml of mixture is taken from the 10-1 dilution and is emptied into the second tube. The second tube now has a total dilution factor of 10-2. 8. The same process is then repeated for the remaining tube, taking 1 ml from the previous tube and adding it to the next 9 ml diluents. 9. As six tubes are used, the final dilution for the bacteria/cells will be 10-6 (1 in 1,000,000).

Some unicellular protists and bacteria, like Thiomargarita namibiensis, are macroscopic and visible to the naked eye. Since the size is a bit of theme in microbiology, let us talk about actual measurements. How small is small? The cellular microbes are typically measured in micrometres (µm). A typical bacterial cell (let us say E. coli) is about 1 µm wide by 4 µm long. A typical protozoal cell (let us say Paramecium) is about 25 µm wide by 100 µm long. Acid Fast Staining It was first developed by Paul Ehrilch in 1882. Dyes like melachite green and methylene blue are used. When smear are treated these dyes and washed with acids and alcohols they are not decolorized and retain stain of dye. Such bacteria which are not decolorized are known as acid fast bacteria. Bacteria which lose stain and got decolorized are known as non acid fast bacteria. Microbiology Notes By Sangita Mam Page 11 Ziehl-Neelsen staining The Ziehl-Neelsen staining technique is a differential staining technique that was initially developed by Ziehl and modified later by Neelsen, hence the name Ziehl-Neelsen stain. 1. On a clean sterile microscopic slide, make the smear of the sample culture and heat fix the smear over blue heat. 2. Over the smear, pour and flood the smear with carbol fuschin and heat gently until it produces fumes. 3. Allow it to stand for 5 minutes and wash it off with gently flowing tap water. 4. Add 20% sulphuric acid and leave it for 1 minutes. Repeat this step until the smear appears pink in color. 5. Wash off the acid with water. 6. Flood the smear with methylene blue dye and leave it for 30 sec and wash with water. 7. Air dry and examine the stain under the oil immersion lens.
